It is a
chromatography technique that is particularly applicable to high molecular mass
species. It is based on different diffusion and permeation rates as different
molecules pass through a porous medium. The fillers are composed of small size
(~ 10 mm) silica or polymer compounds containing a smooth pore network into
which solute and solvent molecules can diffuse. Molecules in the pores of the
fillers are effectively captured and removed by the mobile phase. The duration
of its residence in the pores depends on the size of the molecule. While large
molecular weight compounds exit the column more quickly, smaller molecules
remain in the pores more so they are eluted last.
